An en-suite bathroom is simply a bathroom that is directly connected to the master bedroom. These are a very nice feature, but can be awkward if you only have one bathroom and you have guests over who need to walk through your bedroom to get to the bathroom.

It means that the bathroom is adjoining the bedroom. In other words you don't have to go outside of the bedroom to use the bathroom.
