It could be the result of too much contact with something. If you wear tight pants, pantyhose, or underwear that rubs the same area over and over, it could cause blistering. And there is always the possibility, if you are sexually active, of STDs being the cause.

Some kind of a local infection may cause a blister on your upper thigh. If its a cluster of blisters filled with fluid and painful,then it may be herpes. An insect bite can also cause a blister. If it spreads or persists for more than 4-5 days then please consult a doctor

More than likely an article of clothing you are wearing is tight and rubbing on your upper thigh causing an irritation.
