The first thing to try would be a plunger. If that doesn't work try drain cleaner. Use a thick one meant for removing clogs,(they eat through it) not enzyme cleaners. Make sure the drain cleaner/clog remover is safe for toilets. If all else fails, call a plumber.

The best way is to use a plunger. Plunge until you see all the water drain, and then you know it is safe to flush. If this does not work, you may need to put on some heavy duty plastic gloves and fish out whatever is blocking the toilet's drain. If this fails, call a professional.

If your toilet is overflowing, you can use a plunger and push it into the hole a few times. Then, take it back out and flush the toilet. This should eliminate your toilet dilemma.
